The Quad!!!

Oh yeah!!! Time for you boys and girls to learn about the quad streak, oh yeah, or as I call it, THE QUAD!!!


Now I know what you're thinking, oh yeah, how could the Macho Man Randy Savage know anything about science, MUCH LESS WHAT QUAD STREAKING IS, well let me tell ya. The Macho Man Randy Savage has more to offer than just FLEXING BICEPS AND WANTON VIOLENCE. Oh yeah, you can't become a SIX TIME Heavyweight champion of the World by being a dummy, and Macho Man Randy Savage is no dummy.


Now I'm gonna give it to you real simple yeah. QUAD STREAKING in some ways is just another form of serial dilutions. Scientists use it to create isolated colonies from a mixed group of microorganisms. Oh yeah, just think of it all happening on one plate, as opposed, to multiple plates. Oh yeah, think of each streak as a dilution of 10^-1, to 10^-2, to 10^-3, to 10^-4. Yeah, each streak you do dilutes your inoculum more and more until that LAST STREAK, which should give you the colonies that you need for your experiment. Oh yeah, not that difficult right? Well, let me bring in my tag team partner, Stone Cold Steve Austin, in to finish the job. Oh yeah, when he's done with you, QUAD STREAKING is gonna be easier than snapping into a Slim Jim.


Why thank ya Macho Man. Now, Stone Cold Steve Austin is gonna teach y'all how to perform the quad streak:


First, y'all need to gather your supplies, you're gonna need:

  • An inoculation loop (What?!)

  • Your library plate (What?!)

  • A fresh agar plate (What?!)

  • And a Bunsen burner (What?!)

Now, you're gonna perform the quad streak:

  • You're gonna flame your loop (What?!).

  • You're gonna wait for it too cool (What?!).

  • You're gonna collect a colony from your library plate (What?!).

  • Then create a streak from the edge of the plate towards the center, four lines should be good.

  • Then you're gonna flame your loop again (What?!), then take that loop and create another streak starting from your first streak.

  • To complete the last two streaks of the quadrant, you simply flame your loop again (What?!).

  • Then you create another streak starting from the most recent streak. Then you put it in the incubator at 26 degrees Celsius for 48 hours so your colonies can grow.


Now when Stone Cold does it, I always orient my plate so that my first streak is at the top of the plate. I then turn the plate 90 degrees to the left after each streak so I'm always going from left to right, it helps me not get confused about which streak is which. Other than that, you just need to make sure the loop has cooled down properly. I usually wait 15-20 seconds after I flame the loop to make sure it's Stone Cold enough that it won't kill all the bacteria the moment the loop touches it.


Pretty simple right? All in all, quad streaking just requires a little bit of, flamin (What!). A little bit of, coolin (What!). And a little bit of, streakin (What!). And that's the bottom line cause Stone Cold said so!
